Objectives:We compared the efficacy of three dosage regimens of sildenafil citrate in the treatment of pulmonary arterial hypertension (PAH) in randomized, double-blind, placebo-controlled trials. The efficacy of the first regimen was not different from that of the second and third, although the clinical efficacy in PAH was different. The third regimen did not produce a difference in the incidence of adverse events. The two-drug regimen (Viagra® and Sildenafil®) produced similar safety results. The incidence of adverse events was similar in the three treatments, but not in the one-drug regimen (P = 0.08). The adverse events in the third treatment were mainly mild, with most being reported as an improvement in the patient's condition.

Methodology:The efficacy of the treatment was compared to a control therapy using sildenafil citrate. Patients were randomized to receive one of three sildenafil citrate dosage regimens. The primary outcome was the incidence of adverse events for the three treatment regimens. The efficacy of the first dosage regimen was compared to the control therapy. The incidence of adverse events in the three treatment regimens was statistically equivalent with a treatment difference of only 0.5%, but no treatment difference was observed for the second and third treatment regimens.
